Reconstitution of DNA fragments on HAC/MAC via the fragment-assembly system

A fragment-assembly (FA) system was developed to reconstitute multiple DNA fragments on human and mouse artificial chromosomes (HACs/MACs), allowing simultaneous loading of several PCR-based gene-loading vectors (GLVs) with backbone removal; researchers demonstrated reconstitution of two distinct gene expression cassettes on a MAC, successful transfer of the gene-loaded MAC to other cells, and sustained expression of the introduced genes, with sequences deposited in DDBJ (LC884821–LC884827).
