Novel Mechanism for Parkinson’s Is Linked to ATP Deficiency

Ludwig Maximilian University of Munich researchers report in Science Advances that DJ-1 loss causes ATP deficiency in human iPSC-derived dopaminergic neurons, reducing VMAT2 copy number per vesicle and impairing vesicular dopamine loading. The study links ATP-dependent vesicle uncoating (clathrin accumulation) to dopamine oxidation and shows ATP supplementation rescues defects in vitro, while clinical translation remains distant.
