ABE9 fused to SpRY Cas9 nickase enables precise generation of bystander free mouse models

Researchers report ABE9-SpRY, an adenine base editor fused to a PAM-flexible SpRY-Cas9 nickase, achieves up to 96% precise A-to-G edits with fewer off-targets in mouse embryos and hiPSCs, enabling bystander-free point-mutation models; plasmids are available on Addgene and sequencing data are deposited under SRA PRJNA1291991.
